You need to set appropriate permissions on the new location.
F: -- NETWORK SERVICE needs READ access to the drive
and you need to match permissions for these folders from the original location:
F:\WSUS F:\WSUSContent
among others, NETWORK SERVICE needs WRITE permissions to the ~\WSUSContent folder, subfolders, and files.
The requisite filesystem permissions are documented in Appendix D of the WSUS Operations Guide.
