If the primary does not react in specific time it switches to secondary. Have a look here: http://technet.microsoft.com/en-us/library/cc779517.aspx
If DNS server list is reprioritized, the DNS Client service resets the server priority at periodic intervals. By default, the server priorities are reset every 15 minutes. For XP see here also: http://support.microsoft.com/kb/320760/en-us
You can use nslookup for the used server and lot of other queries: http://support.microsoft.com/kb/200525
